UV-1280 and Water Analysis Program with Sipper-160L

簡介

UV-1280是目前島津唯一一臺single beamUV 分光光度計。效能當然比其他double beam的機種差,新的LabSolutions UV也不支援。如果要用電腦控制,也只能用舊的UVProbe。優點當然是便宜,體積也是部份原因。

不過有個選購項目Water Analysis Program。這是一整套解決方案,可以對22種水樣測試39個測項。只要將水樣加入細川株式會社 Kyoritsu Chemical-Check Lab., Corpkit處理,然後直接送入儀器中即可測試,以內建檢量線計算出濃度。

Instroduction

UV-1280 is the only single beam UV spectrometer of Shimadzu.  Performance is not as good as other double beam spectrometers, and LabSolutions UV-vis doesn’t support it.  If you still want to control it by computer, the only option is the old UVProbe.  The benefit is cheaper, and smaller size. 

There is a option “Water Analysis Program” for UV-1280.  It’s a complete solution for analyzing 22 sample types/39 items.  Just pretreat water sample into the kit provided by Kyoritsu Chemical-Check Lab., Corp, load into UV-1280.  The instrument will quantify the water sample for you.

This program is already embedded in UV-1280 firmware, just needs an authentication key to activate it.  Insert it into any USB port on rear panel of UV-1280.

軟體操作  Software Operation

開機後,UV-1280會自動執行自我檢查。如果一切正常,會進入模式選單。

選擇  9. Option1. Water Analysis,就會進入Water Analysis 的選單

UV-1280 will perform initialization after powering on, and enter Mode Menu Screen if everything is ok.

Pick 9. Option, 1. Water Analysis, you can access Water Analysis Program menu (Measurement Item Selection screen).





 



在這裡可以按左右鍵換頁,可以看到不同的測項。手冊以選擇Cr6(WAK),六價鉻(WAK)為例:

進入這個畫面可以看到,儀器自動設成所需的波長,1.5mL Cell是指semi-micro cell。同時也告訴你要用哪一個試劑去處理樣品。這裡寫的量測步驟不是很清楚,只能當作提示。詳細的步驟可參考手冊的5.3 Measuring method of each item

1.      將未處理的樣品放入cell,按CellBLK(F3) 測量背景值。

2.      1.5毫升樣品加入杯中,液面應對齊樣品杯上的刻線

3.      將所有樣品吸入試劑管中並立即按下Measure

4.      立即搖晃試劑管五六次,將樣品倒入cell,將cell置入UV-1280

5.      按下Measure後,計時器會開始倒數計時。以這個測項為例,兩分鐘。之後儀器會開始測量,並直接顯示結果(0.871)

6.      必須在樣品吸入試劑管後兩分鐘量測。如果沒有及時按下Measure,必須在兩分鐘時按下Manual測量(0.770)。要注意的是,要在兩分鐘到之前按下SaveData把結果存檔。如果兩分鐘到了,儀器量測出結果,那就只能存這個結果。

7.      換句話說,如果太早按[Measure],之後才將樣品吸入試劑管,UV-1280就會太早測量而提供錯誤的結果,因為反應時間不足。

We can find instrument switches to the wavelength for this item.  This screen assigns a 1.5mL cell (semi-micro) and regent to pretreat sample.

1.      Load untreated sample into cell, press CellBLK for background.

2.      Add 1.5mL sample in a specific cup.  Meniscus should be aligned to the line.

3.      Suck all sample into the pack test tube, and press [Measure] at the same time.

4.      Shake and mix the tube 5 or 6 times, and load the mixed sample into a cell, load the cell into UV-1280.

5.      After pressing [Measure], the timer is counting down.  In this case, 2 minutes.  After it, UV-1280 will measure the sample and display the result (0.871).

6.      You must measure the sample after sucking sample into pack test tube at two minutes sharp.  If you didn’t press [Measure] in time, you should press [Manual] at 2 minutes to measure the sample (0.770).  Note: you must save the manual result by pressing [SaveData] before wait time elapsed.  After UV-1280 measured it, you can only save the result (0.871).

7.      In other words, if you press [Measure] too early, and suck sample into tube late, UV-1280 will measure the sample too early and provide incorrect result because there is not enough reaction time.

我不確定這是什麼的縮寫UV-1280可以選配這五種樣品模組,不過Water Analysis Program只支援前三種。不過我還是可以用Sipper 160。我不確定Syringe sipper到底行不行,要有機會測試。

·        Standard Cell沒什麼好講,換樣品就是換裝一個cell

·        6-cell CPS (其實都是六個,只是CPS可控溫,另一個不行)要進入SmptCmpt中換到要測量的cell位置。

I’m not sure what this abbreviation means.  UV-1280 is competable with these five sample modules, but Water Analysis Program only supports the first three.  I can still use Sipper 160, with some restrictions.  I’m not sure if Syringe sipper works or not since I don’t have opportunity to test it.

·        Standard Cell: just load next cell for next sample.

·        6-cell and CPS: They both holds 6 cells, CPS can control temperature but the other doesn’t.  You have to enter “SmptCmpt” screen to switch to desired cell position.







For using Sipper 160 (Just leave Syringe Sip screen here for later use)

·        必須在進入Water Analysis Program 之前選擇Sipper 160

o   必須啟動ManualSip

·        測量每樣品都必須進入SmptCmpt畫面控制pump 抽取樣品。

·        能用按蠕動幫浦的蓋子來啟動或停止幫浦。幫浦會照設定的轉速運轉,但是其他的設定通通沒用。

·        You have to select it and set up parameters before entering Water Analysis Program.

o   ManualSip has to be enabled.

·        You have to enter this screen to sip each sample.

·        Start/stop peristaltic pump by pressing the lever.  Pump works at the setting speed, but all other settings are ignored.

我的意見  My comments

1.      不用自己作檢量線的確非常方便。只是有的實驗室可能無法接受,結果也可能有少許誤差。

2.      有的測項的樣品太少,1.5毫升應該沒辦法用sipper

3.      6-cell holder好像沒甚麼用。樣品必須在規定的等待時間測量,不能先準備好三組一起放進去。

4.      Manual Measurement好像也沒什麼用,因為我還是必須計時。如果我加了試劑但忘了按Measure,因為沒計時,我也不知道何時該測量。如果太早按,測出來的值也是不能用。所以,如果沒有在樣品混合試劑時同時按下Measure,就直接認命重作。

5.      除了Standard Cell之外,要進入SmptCmpt畫面去換每樣品實在相當不方便。

 

1.      It’s very convenient that you don’t have to make calibration curves.  The problem is some labs may not accept it, and there might be some errors.

2.      There are several tests that don’t have enough sample volume because of the pretreatment method.  It’s not possible to use Sipper if the sample volume is only 1.5mL.

3.      6-cell holder doesn’t help.  You must measure the sample at a specific time according to the reaction, you cannot prepare three samples and load them together.

4.      Manual Measurement doesn’t help.  I still need to set a timer to know the correct reaction time to press [Measure].  You don’t know you should set up a timer when you press it too late, and the result is not acceptable if you press it too early.  If you didn’t press [Measure] when mixing reagent with sample, re-prep this sample.

5.      You have to enter SmptCmpt screen to load each sample other than standard cell.  It’s very inconvenient.
